Detective Naani opens with a regular day at Gulmohar Complex where the spunky and independent 75-year-old Naani (Ava Mukherjee) is on her way back home from her daily morning walk. Suddenly, her eye catches the face of a little girl peeping nervously from a third floor window of a flat belonging to a newly arrived childless couple. The girl moves away quickly and Naani is intrigued by this little girl.
She decides to pursue her hunch and starts an amateur investigation on this. First she goes to the CID who dismiss her story due to the lack of hard evidence and then she summons up her own detective squad. This comprises of her dog Bruno, 2 inquisitive little grandchildren Nakul (Zain Khan) and Anjali (Simran Singh), her divorced daughter Priya(Amrita Saluja), teenagers Neeti (Shweta Gulati) and Rohan (Amit Varma), and the elderly Mr.Pal (Hemu Adhikari) who serves as Naani's binocular-toting ground floor spy. Clue after clue leads Naani to a racket where the stakes are high, the criminals ruthless and the leader, very powerful.
Naani, unfortunately, gets too close to the trail and endangers herself. Soon she finds herself sharing the same plight as the little girl. Her detective squad gets together in an attempt to rescue Naani with a little help from the CID. On the way, a romance blossoms between the squabbling teenagers and the divorced daughter and the dashing CID inspector. Some humour, suspense and a bit of insight peppers the story on its way.
